Proteins carry out the basic functions of complex biological systems and can serve as enzymes, binding partners, or cellular ultrastructures. Characterization of differential protein expression is commonly achieved via specific immunodetection (western blot analysis) of biological samples. Historically, sensitive detection of proteins has utilized HRP-linked secondary antibodies and chemiluminescent substrates. Use of fluorescent secondaries, however, allows efficient assessment of multiple proteins simultaneously on a single western blot. This webinar will outline protocols for fluorescent immunoblots, focusing on the multiplex analysis of complex samples. We will also discuss recent advancements in fluorophores used in western blotting and multiplex image acquisition.
